iGEM Dundee 2014

This cblD promoter region from Burkholderia cenocepacia was hooked up to a GFP gene from Dundee iGEM 2011 BBa_K562012. This was then co-expressed with Burkholderia BCAM2207 (a synthetic gene that could not be BioBricked at this time) and BCAM2208 from BBa_K1315007 in an E. coli chassis (Dundee Figure 1). The device was found to produce GFP without exposure to the normal inducer molecule, BDSF (Dundee Figure 1). Interestingly, the cblD promoter seems to be activated by BCAM2208 even without the sensor kinase (Dundee Figure 2). Thus the response regulator/promoter is constitutively active in an E. coli chassis.
